fa تاثیر تمرینات استقامتی و مصرف گالیک اسید و کامپفرول بر بیان ژنهای ARC، VEGFα ،HIF1α وCREB1 در موش های مبتلا به سرطان پستان تحت درمان با داروی پاکلیتاکسل The effect of endurance training and consumption of gallic acid and kaempferol on the expression of ARC, VEGFα, HIF1α and CREB1 genes in mice with breast cancer treated with paclitaxel تغذیه، روانشناسی، ورزش Nutrition, psychology, sport پژوهشي Research <div style="text-align: justify;">مقدمه: سرطان پستان شایع‌ترین سرطان است. استراتژی‌های درمانی سبب کاهش نورونزایی در بیماران تحت درمان می‌شود. هدف تحقیق حاضر، بررسی تأثیر تمرینات استقامتی و مکمل گالیک اسید و کامپفرول بر نورون‌زایی در هیپوکمپ، ناشی از عوارض جانبی مصرف پاکلیتاکسل در موش‌های مبتلا به سرطان پستان است. روش بررسی: بطور تصادفی موش‌ها به شش گروه (6=n) شامل: گروه کنترل، گروه مبتلا به سرطان پستان با تزریق رده سلولی 4T1، گروه سرطانی که داروی پاکلیتاکسل (10 میلی‌گرم بر کیلوگرم) دریافت شد، گروه سرطانی تحت تیمار با پاکلیتاکسل و گاواژ مکمل کامپفرول و گالیک اسید، گروه سرطانی تحت تیمار با پاکلیتاکسل و تمرین استقامتی، گروه سرطانی تحت تیمار با پاکلیتاکسل و تمرین استقامتی و مکمل تقسیم شدند. بیان ژن‌ها به روشReal Time-PCR سنجیده شد. ژن‌ها به کمک نرم افزارهای بیوانفورماتیک انتخاب شدند. تجزیه و تحلیل آماری توسط نرم‌افزار GraphPad-Prism نسخه 9 انجام شد. داده‌ها با استفاده از آنالیز واریانس یک‌طرفه (ANOVA) با آزمون Tukey's post hoc محاسبه گردید. نتایج به‌عنوان میانگین±انحراف استاندارد نشان داده شد و P.value<0.05 نشان‌دهنده معنی‌داری است. یافته‌ها: بیان VEGFα/ARC/CREB در اثر مصرف گالیک اسید، کامپفرول و تمرینات استقامتی در موش‌های شیمی درمانی شده افزایش یافت (0/05p<). همچنین، بیان VEGFα/ARC/CREB در گروه تمرین استقامتی همراه با مصرف مکمل افزایش یافت (0/05p<). سرطان پستان سبب افزایش بیان HIF1α شده است (0/05p<). تمرین استقامتی و مصرف مکمل میزان بیان HIF1α را بطور معنی‌داری کاهش داده است (0/05p<). نتیجه‌گیری: تمرین استقامتی و مصرف گالیک اسید و کامپفرول سبب تنظیم بیان ARC، VEGFα ، HIF1α وCREB1 در بافت مغز موش‌های مبتلا به سرطان پستان می‌گردد.  </div> <div style="text-align: justify;">Introduction: Breast cancer is the most common cancer. Therapeutic strategies reduce neurogenesis in treated patients. The present study aimed to assess the effect of endurance training along with the combination of gallic acid and Kaempferol on neurogenesis in the hippocampus caused by the side effects of paclitaxel in mice with breast cancer. Methods: The mice were randomly assigned to six groups (n=6), including control group, the group with breast cancer injected with 4T1 cell line, the cancer group that received paclitaxel (10 mg/kg), cancer group treated with paclitaxel and gavage supplemented with Kaempferol and gallic acid, cancer group treated with paclitaxel and endurance exercise, the cancer group treated with paclitaxel and endurance training and supplements. Gene expression was measured by the Real Time-Polymerase Chain Reaction (PCR) method. Genes were selected using bioinformatics software. In this study, statistical analysis was performed using GraphPad Prism software (version 9). Data were calculated using one-way analysis of variance (ANOVA) with Tukey's post hoc test. Results were presented as mean±standard deviation. A p-value less than 0.05 was considered statistically significant. Results: The expression of VEGFα/ARC/CREB increased significantly (P<0.05) due to the consumption of gallic acid, kaempferol, and endurance training in mice treated with paclitaxel. Moreover, the expression level of VEGFα/ARC/CREB genes increased in the endurance training group and the supplement (P<0.05). On the other hand, breast cancer has increased the expression of HIF1α gene (P<0.05). Endurance training and supplement consumption significantly reduced the expression of the HIF1α gene (P<0.05).
