مقدمه: فناوری نانو از طریق طراحی سیستم‌های دارورسانی هوشمند، چالش دارورسانی هدفمند و اسیدهای نوکلئیک به سلول‌های سرطانی را برطرف می‌کند. این رویکرد، دقت و کارایی بهبود یافته در دارورسانی عوامل درمانی را ممکن می‌سازد. این مطالعه بر طراحی و ارزیابی یک نانوحامل چندمنظوره مبتنی بر PLA/Chitosan/PEG عامل‌دار شده با اسید فولیک (FA) با قابلیت هدف‌گیری تومور و دارورسانی هم افزایی پاکلی­تاکسل (PTX) و siRNA به سلول‌های سرطان سینه (MCF-7) تمرکز دارد که هدف آن افزایش جذب انتخابی و کارایی درمانی در مقایسه با سیستم‌های دارورسانی مرسوم است.
روش بررسی: سنتز نانوذرات (FA/PLA/Chitosan/PEG) PCPF به کمک روش پیونددهی شیمیایی و فرآیند حل-در-حلال انجام شد. سپس دارو PTX و siRNA برعلیه ژن  Survivinبا روش انتشار حلال همراه با امولسیون چندمرحله‌ای در نانوذرات PCPF بارگذاری شدند. ویژگی­های فیزیکوشیمیایی و مورفولوژی نانوذرات و نانوکپسول ­های سنتز شده از طریق 1H-NMR، FTIR و DLS بررسی شد. پس از بررسی تأثیر­pH های مختلف (5 و 4/7) بر رهایش محموله‌ها، سمیت سلولی نانوکپسول‌ها با آزمون MTT و آپوپتوز با روش Annexin V/PI در رده‌های MCF-7 و MCF-10A ارزیابی شد. آنالیز آماری داده‌ها با استفاده از ANOVA یک‌طرفه و آزمون دانکن (SPSS 22، سطح معنی‌داری 5 درصد) انجام شد.
یافته ­ها: نتایج آنالیز 1H-NMR و FTIR تشکیل موفقیت‌آمیز نانوذرات PCPF را تأیید کرد. طبق نتایج حاصل از DLS مشخص شد که اندازه نانوذرات از 6±162 نانومتر برای  PCPFتا 11±256 نانومتر برای PCPF/siRNA/PTX  متغیر بود. هم­چنین مشخص شد که بارگذاری PTX و siRNA اندازه و PDI نانوذرات را افزایش داد و پتانسیل زتا عمدتاً منفی مشاهده شد. مطالعات رهایش، نشان داد رهایش PTX و siRNA در pH  اسیدی (5) به‌طور قابل‌توجهی بیشتر از pH خنثی (4/7) بود. آزمون ­MTT، سمیت انتخابی نانوذرات حاوی PTX و siRNA را علیه سلول‌های سرطانیMCF-7 و کاهش عوارض بر سلول‌های سالم MCF-10A در مقایسه با داروی آزاد، را اثبات کرد. هم­چنین نتایج آپوپتوز سلولی نشان داد که بیشترین میزان پیش و پس‌آپوپتوز سلولی (به ترتیب 32/25 و 85/19 درصد) برای سلول‌های MCF-7 مربوط به نانوکپسول‌های PCPF/siRNA/PTX بود، در حالی که بیشترین درصد سلول ­های واقع در مرحله نکروز (59/30 درصد) مربوط به PTX آزاد بود.
نتیجه ­گیری: به‌طور کلی، نانوذرات PCPF مبتنی بر PLA-PEG و هدفمندشده با فولیک‌اسید، با رهش کنترل‌شده وابسته به pH و زیست‌سازگاری مطلوب، سمیت انتخابی و القای غالب آپوپتوز را نشان داده و از طریق انتقال همزمان PTX/siRNA، به‌طور معنی‌داری موجب افزایش اثر سینرژیستی درمانی در سلول‌های سرطانی شدند.
