Introduction: Mesenchymal stem cells (MSCs) can modulate the tumor microenvironment and influence cancer cell behavior by secreting various bioactive factors. This study aimed to investigate the effect of MSC-derived secretions from chick embryo lung tissue on the expression of the tumorigenic genes WNT4 and GLI2 and of inflammatory factors in a human breast cancer cell line (MCF-7).
Materials and Methods: In this in vitro experimental study, cells were isolated and cultured from 10-day-old chick embryo lung tissue. In the third passage, the supernatant (CM; conditioned medium) obtained from MSCs was collected, and its effect on MCF-7 cells was examined. After 48 hours, CM was collected and filtered. ELISA measured the levels of the inflammatory cytokines IL-6 and TNF-α, and qRT-PCR measured the levels of the WNT4 and GLI2 genes. For statistical comparisons between groups, an independent t-test was used. All analyses were performed using SPSS version 26, and 0.05 was considered the significance threshold.
Results: Treatment of MCF-7 cells with MSCs-CM resulted in a significant decrease in the expression of WNT4 (P=0.0034) and GLI2 (P=0.0007) genes in the treatment group compared to the control group. Also, the levels of IL-6 (P=0.0155) and TNF-α (P=0.0073) in the treatment group showed significant decreases.
Conclusion: MSCs-CM can exert inhibitory effects on cancer cells by downregulating key genes in tumorigenic signaling pathways and reducing inflammatory factors.
