Introduction: Breast cancer is one of the major public health challenges, and the search for natural-origin assistive therapies has become increasingly important. The study aimed to assess the therapeutic effects of the essential oil of Echinophora cinerea (EC-EO) in the treatment of breast cancer using a cell line and a rat model with a 7,12-dimethylbenz [a]anthracene (DMBA)-induced tumor.
Methods: A hybrid essential oil (70% leaves and 30% stems) of the collected plant was extracted, and its phytochemical compounds and antioxidant potential were evaluated. Cell toxicity on the MCF-7 hormone-dependent cell line was examined. In the in vivo phase, rats with induced tumors received oral gavage doses of 50 and 100 mg/kg EC-EO for 6 weeks. Tumor parameters, histological changes, and blood indicators were compared with those of the control groups.
Results: The essential oil is rich in terpenoids, with predominant Alpha-phellandrene and p-Cymene compounds. The extract exhibited strong antioxidant activity and dose-dependent cell toxicity against MCF-7 cancer cells. In the animal model, the high-dose-treated group showed a significant decrease in tumor weight (55.6% inhibition) and tumor survival rate (50%) compared with the negative control group (p < 0.001). Also, significant improvements in hematological parameters and a reduction in tissue lesions (decreased cell density, necrosis, and invasion) were observed.
Conclusion: The essential oil of Echinophora cinerea, with its rich phytochemical content, exhibits significant anti-tumor and therapeutic effects that depend on the dose. This compound can be considered a natural auxiliary therapeutic agent in the control of breast cancer.
