Introduction: The tumor microenvironment and factors secreted by adjacent cells play a crucial role in the progression or inhibition of cancer. This study was designed to investigate the effect of supernatant from chick embryo hepatocytes on the protein levels of inflammatory (IL-6) and anti-inflammatory (IL-10) factors as well as the expression of cell cycle-related genes in a breast cancer cell line (MCF-7).
Methods: In this in vitro experimental study, hepatocytes were isolated from chick embryo livers on the 10th day of incubation and cultured in DMEM/F12 medium containing 10% fetal bovine serum (FBS). After 48 hours, the supernatant was collected and filtered. MCF-7 cells were divided into two groups: control (treatment with basal medium) and intervention (treatment with supernatant) and incubated for 48 hours. IL-6 and IL-10 cytokine levels in the supernatant of MCF-7 cells were assessed using ELISA and relative expression of cyclinD1 and P53 genes was assessed using Real-Time PCR. Data analysis was performed using SPSS version 26 software and independent t-test, and a significance level of 0.05 was considered.
Results: Treatment of MCF-7 cells with hepatocyte supernatant significantly increased the protein levels of IL-10 (P=0.015) and IL-6 (P=0.013) in MCF-7 cells. Also, in these cells, P53 expression increased (P=0.002) and Cyclin D1 expression decreased (P=0.045) significantly.
Conclusion: Hepatocyte cell supernatant can exert regulatory and potential antitumor effects on MCF-7 cell line by inducing IL-10 expression, activating P53-dependent pathway and inhibiting Cyclin D1 expression. However, the concomitant increase in inflammatory factor IL-6 requires further mechanistic studies.
